The 5-HT7 receptor is primarily found in the CNS, GI tract, and blood vessels. Serotonin is the primary ligand of the 5-HT7 receptor, which activates G(s) proteins associated with the receptor and leads to the activation of adenylyl cyclase and production of the secondary messenger cyclic adenosine monophosphate (cAMP). cAMP activates protein kinase A, phosphorylating downstream effectors that lead to specific cellular responses which mediate physiological functions such as thermoregulation, circadian rhythm, learning and memory, sleep, and potentially mood.
