Methotrexate is a chemotherapeutic drug and is a folic acid derivative that inhibits many enzymes involved in nucleotide synthesis. Methotrexate travels across tissues and inhibits dihydrofolate reductase, thymidylate synthase and aminoimidazole carboxamide ribonucleotide transformylase (AICRT), without these enzymes nucleotide synthesis cannot occur preventing cell division. Methotrexate is metabolized by folypolyglutamate synthase into methotrexate polyglutamate, which inhibits AICRT more than methotrexate, this leads to the accumulation of adenosine triphosphate and activation of the anti-inflammatory cascade. Some overdose symptoms that can occur include nausea, vomiting, gastrointestinal issues and or complications with red blood cells, white blood cells and or platelets. When taking this drug avoid substances such as alcohol, dairy products, St.John's wort and caffeine.
