Dalfampridine, also known as Ampyra (USA) and Fampyra (Canada and Australia), is a potassium channel blocker. This drug is used for the improvement of motor function in patients with multiple sclerosis (MS). Dalfampridine is a board-spectrum lipophilic potassium channel blocker and binds favorably to the open state of the potassium channel in the CNS and in the neuromuscular junctions. Its pharmacological target is the potassium channels exposed in MS patients because their axons are progressively demyelinated. As a result, there is a leak of potassium ions which results in the repolarization of cells and a decrease in neuronal excitability in the neurons of those patients. The impact is the impairment of neuromuscular transmission as it is harder to trigger an action potential in the neurons. Dalfampridine has the function to make sure that the current available in the neurons is high enough to stimulate conduction even in demyelinated axons that are exposed in MS patients. Dalfampridine is administered as an oral tablet.
