Galantamine is a cholinesterase inhibitor used in the treatment of Alzheimer's disease, specifically to mitigate symptoms of mild to moderate dementia. These symptoms of cognitive loss are due to acetylcholine deficiency, galantamine inhibiting acetylcholinesterase and cholinesterase, which increases extracellular levels of acetylcholine. It also acts as an allosteric modulator for the neuronal acetylcholine receptor, helping control the release of neurotransmitters like acetylcholine, GABA, glutamate, dopamine, serotonin and norepinephrine ultimately aiding in performance in cognitive tasks. The drug is metabolized by enzymes CYP2D6 and CYP3A4 into metabolites. In case of an overdose atropine should be administered intravenously in order to counter the effects of galantamine by a trained professional.
